Biological Background: Thiosulfate Sulfurtransferase Function and Localization
- Thiosulfate sulfurtransferase (TST), also known as rhodanese, is a mitochondrial enzyme involved in sulfur transfer and cyanide detoxification.
- Catalyzes the transfer of a sulfur ion from thiosulfate to cyanide, forming thiocyanate; a key reaction in cyanide clearance (PubMed:40033006). Also exhibits weak mercaptopyruvate sulfurtransferase (MST) activity (By similarity). Related references: PMID:40033006
- Together with MRPL18, acts as a mitochondrial import factor for the cytosolic 5S rRNA; only the nascent unfolded cytoplasmic form binds 5S rRNA (PubMed:20663881, PubMed:21685364). Related references: PMID:20663881 PMID:21685364
- Localized to the mitochondrial matrix.
- Post-translationally modified by acetylation and phosphorylation; also reported as a glycoprotein.
Experimental Guidance and Technical Tips
- The immunogen encompasses nearly the entire human TST sequence (aa 1–297), enabling recognition of multiple epitopes in denaturing and native assays.
- For Western blotting, a single band at ~33 kDa is anticipated; ensure mitochondrial protein extraction protocols are used.
- In immunohistochemistry (paraffin), antigen retrieval conditions may need optimization for mitochondrial targets.
- In immunoprecipitation, the antibody may capture native TST; validate in your sample system.
- For ELISA, coat with recombinant TST protein and employ standard polyclonal detection workflows.
