Biological Background: Thioredoxin 1 (Trx1/TXN) Function and Localization
- Thioredoxin 1 (TXN), also known as ATL-derived factor (ADF) or surface-associated sulphydryl protein (SASP), is a multifunctional redox-active protein encoded by the TXN gene.
- Catalyzes dithiol-disulfide exchange reactions through reversible oxidation of its active center dithiol to a disulfide. Related references: PMID:17182577 PMID:19032234 PMID:2176490
- Mediates reversible S-nitrosylation of cysteine residues in target proteins; nitrosylates the active site cysteine of CASP3, inhibiting caspase-3 activity in response to nitric oxide. Related references: PMID:16408020 PMID:17606900
- Induces FOS/JUN AP-1 DNA-binding activity and stimulates AP-1 transcriptional activity in ionizing radiation-treated cells. Related references: PMID:11118054 PMID:9108029
- ADF augments the expression of the interleukin-2 receptor TAC (IL2R/P55).
- Localizes to the nucleus, cytoplasm, and can be secreted from cells.
- Post-translational modifications include acetylation, disulfide bond formation, S-nitrosylation, and ubiquitination; serves as an electron transport and transcription regulation factor.
Experimental Guidance and Technical Tips
- The immunogen, a synthetic peptide spanning amino acids 26-105 of human TXN, targets a conserved region across human, mouse, and rat, enabling the observed cross-reactivity.
- For Western blot (WB), the predicted molecular weight is approximately 12 kDa; consider using a 12-15% gel and optimizing transfer conditions for low molecular weight proteins.
- In immunohistochemistry (IHC-P), ensure proper antigen retrieval and validate staining patterns in relevant tissue samples to minimize non-specific binding.
- For ELISA, the antibody can be used as a capture or detection reagent; titrate to determine optimal working concentration in your assay system.
