- TGS1, also known as PIMT, PIPMT, or NCOA6IP, is a trimethylguanosine synthase that catalyzes the two-step hypermethylation of the 7-monomethylguanosine (m7G) cap of snRNAs and snoRNAs to a 2,2,7-trimethylguanosine (m2,2,7)G cap structure.
- The enzyme is specific for guanine and requires N7 methylation before N2 methylation, using S-adenosyl-L-methionine as a methyl donor.
- Hypermethylation of the m7G cap by TGS1 promotes the concentration of U snRNAs in nuclear foci, their colocalization with coilin, and the formation of canonical Cajal bodies (CBs).
- Beyond snRNA processing, TGS1 plays a role in transcriptional regulation as a nuclear receptor coactivator 6-interacting protein.
- Immunofluorescence and subcellular fractionation studies localize TGS1 to the cytoplasm, nucleus, Cajal bodies, and nucleolus.
- TGS1 is ubiquitously expressed with high levels detected in heart, skeletal muscle, kidney, liver, and placenta.
