- Within hair follicles, TGM3-mediated cross-linking of structural proteins is essential for hardening the inner root sheath, contributing to hair strength.
- TGM3 is predominantly cytoplasmic and requires calcium binding for activation; it is synthesized as a zymogen that undergoes proteolytic cleavage to expose the active site.
- The protein is subject to post-translational modifications including acetylation and phosphorylation, and is classified under metal-binding, transferase, and keratinization-related protein families.
