Biological Background: TGF-beta receptor type-1 (TGFBR1) Function and Localization

  • TGFBR1 encodes a single-pass transmembrane serine/threonine kinase receptor that partners with TGFBR2 to form the non-promiscuous receptor for TGF-beta cytokines (TGFB1, TGFB2, TGFB3).
  • Upon ligand binding, TGFBR2 phosphorylates and activates TGFBR1, which then phosphorylates SMAD2; the SMAD2-SMAD4 complex translocates to the nucleus to regulate transcription of target genes, defining the canonical TGF-beta pathway.
