Biological Background: TFF1/pS2 Function and Localization

  • TFF1 (Trefoil factor 1) is also known as breast cancer estrogen-inducible protein (pS2), PNR-2, polypeptide P1.A (hP1.A), commonly referred to as pS2, BCEI, HPS2, HP1.A, pNR-2, and D21S21.
  • It functions as a stabilizer of the mucous gel overlying the gastrointestinal mucosa, providing a physical barrier against noxious agents, and may inhibit the growth of calcium oxalate crystals in urine.
  • The protein is secreted, containing a signal peptide and forming disulfide bonds characteristic of the trefoil factor family.
  • TFF1 is strongly expressed in normal gastric mucosa and in regenerative tissues surrounding ulcerous lesions of the gastrointestinal tract, but lower expression is found in gastric cancer.
  • It is detected in goblet cells of the intestines, submucosal glands of the esophagus, trachea, and vagina, as well as in urine; weakly in bronchial goblet cells.
  • TFF1 is strongly expressed in breast cancer while normal mammary tissue shows low levels; its expression is regulated by estrogen in MCF-7 cells.
  • According to UniProt keywords, it is a growth factor, has a 3D-structure determined, and is identified in proteomics studies.

  • For WB, given the small protein weight of TFF1 (~9 kDa), consider using a 15% SDS-PAGE gel and optimize transfer to low-molecular-weight PVDF or nitrocellulose membranes to ensure retention.
  • When performing IHC-P, validate in known positive tissues such as human gastric mucosa or breast carcinoma, and include appropriate antigen retrieval steps.
