Biological Background: TFF1 Function and Localization

• TFF1 (trefoil factor 1), also known as pS2, BCEI, or PNR-2, is a small secreted protein belonging to the trefoil factor family,
• It functions as a stabilizer of the mucous gel overlying the gastrointestinal mucosa, providing a physical barrier against noxious agents.
• May inhibit the growth of calcium oxalate crystals in urine.
• Predominantly localized in the upper gastric mucosal cells, with additional expression in goblet cells of the small and large intestine, rectum, and scattered expression in respiratory and reproductive tract epithelia.
• Strongly expressed in breast cancer but at low levels in normal mammary tissue; expression is regulated by estrogen in MCF-7 cells.
• High expression in normal gastric mucosa and regenerative tissues surrounding gastrointestinal ulcerous lesions, but lower in gastric cancer.
• Classified as a growth factor and contains a characteristic disulfide bond; annotated as a secreted protein with a signal peptide.
