Biological Background: TFEB Function and Localization
- Transcription factor EB (TFEB), also known as Tcfeb and bHLHe35, is a key member of the MiT/TFE family of transcription factors and functions as a master regulator of lysosomal biogenesis, autophagy, and energy metabolism. Related references: PMID:16936731, PMID:22343943, PMID:27278822
- TFEB specifically recognizes E-box sequences (5'-CANNTG-'), and efficient DNA-binding requires dimerization with itself or other MiT/TFE family members (e.g., TFE3, MITF). - TFEB activity is tightly regulated by MTOR-dependent phosphorylation: in nutrient-rich conditions, phosphorylation retains TFEB in the cytoplasm; upon starvation or lysosomal stress, MTOR inhibition leads to dephosphorylation, nuclear translocation, and transcriptional activation of target genes. - In immunoprecipitation, optimal antibody concentration should be titrated for the target species and sample type; inclusion of appropriate positive and negative controls is recommended.
