Biological Background: TFE3 Function and Localization

  • TFE3 is a transcription factor that dimerizes with itself or other MiT/TFE family members such as TFEB or MITF to bind E-box sequences (5'-CANNTG-3') and regulate gene expression. Related references: PMID:24448649

  • Acts as a master regulator of lysosomal biogenesis by specifically recognizing the CLEAR-box sequence (5'-GTCACGTGAC-3') in promoters of lysosomal genes, thereby activating their expression. Related references: PMID:24448649

  • TFE3 activity is tightly controlled by mTOR-dependent phosphorylation: in nutrient-rich conditions, mTOR phosphorylates TFE3, promoting cytoplasmic retention and inactivation; upon starvation or lysosomal stress, dephosphorylation drives nuclear translocation and transcriptional activation. Related references: PMID:24448649, PMID:31672913, PMID:36608670

  • Maintains the pluripotent state of hematopoietic stem cells by promoting the expression of pluripotency genes; mTOR-dependent cytoplasmic retention of TFE3 promotes exit from pluripotency. Related references: PMID:30733432

  • Recognizes the MUE3 box, a subset of E-boxes present in the immunoglobulin enhancer, and binds to a USF/MLTF site, implicating a role in immune response regulation. Related references: PMID:2338243

  • Promotes lysosomal positioning in response to nutrient deprivation by inducing expression of PIP4P1. Related references: PMID:29146937

  • Subcellular localization dynamically shifts between cytoplasm, cytosol, nucleus, and lysosome membrane depending on nutrient status and mTOR signaling.

  • Post-translationally modified by phosphorylation, methylation, and ubiquitination; also undergoes alternative splicing.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-160) of human TFE3, which may allow recognition of multiple isoforms. For western blot (WB), the predicted molecular weight is approximately 62 kDa; bands near this size should be expected.
  • Validate the antibody in your specific sample system; human, mouse, and rat TFE3 are reported to be reactive. Consider using nuclear extracts or whole cell lysates for detection, as TFE3 shuttles between compartments.
  • For ELISA, optimize coating conditions and antibody dilution using known positive and negative control antigens. Since this is a polyclonal antibody, ensure consistency across batches if used in quantitative assays.
