Biological Background: TFB1M Function and Localization
- TFB1M, dimethyladenosine transferase 1, mitochondrial, is an S-adenosylmethionine-dependent methyltransferase that dimethylates adjacent adenosines A1583 and A1584 in the loop of helix 45 of 12S rRNA, a modification essential for the assembly or stability of the small mitochondrial ribosomal subunit. Related references: PMID:12496758, PMID:25305075, PMID:31251801
- It also plays a role in basal transcription of mitochondrial DNA by interacting with POLRMT and TFAM, and this transcriptional stimulation is independent of its methyltransferase activity. Related references: PMID:11809803, PMID:12068295, PMID:12897151
- The protein is located in the mitochondrion.
- TFB1M is ubiquitously expressed in human tissues.
- It belongs to the methyltransferase family and contains domains for S-adenosyl-L-methionine binding, RNA binding, and DNA binding, functioning in rRNA processing, transcription regulation, and mitochondrial gene expression.
- Alternative names include CGI75, mtTFB, CGI-75, mtTFB1, and h-mtTFB1.
- The precursor protein includes a transit peptide for mitochondrial targeting; the mature enzyme is approximately 40 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 28–346 of human TFB1M, which excludes the N‑terminal mitochondrial targeting sequence; thus the antibody may detect both precursor and mature forms. In Western blot, a band around 40 kDa is expected, consistent with the calculated molecular weight of the processed protein.
- For Western blot, mitochondrial‑enriched fractions from human, mouse, or rat cell or tissue lysates may provide optimal detection. Validate the antibody in your specific sample type and lysis conditions.
- The antibody is qualified for ELISA; consider using the recombinant immunogen as a positive control when designing assays.
- Due to TFB1M’s dual roles in mitochondrial rRNA modification and transcription, functional studies (e.g., methyltransferase activity assays or transcription reporter systems) can complement detection by western blot.
