Biological Background: TFAM Function and Localization
- Transcription factor A, mitochondrial (TFAM, also known as MtTF1, TCF-6, TCF6L2) is a key regulator of mitochondrial DNA (mtDNA) transcription and maintenance.
- Binds to the mitochondrial light strand promoter to regulate transcription initiation; essential component of the mitochondrial transcription initiation complex with TFB2M and POLRMT. Related references: PMID:29149603
- Promotes transcription from both the heavy strand promoter 1 (HSP1) and the light strand promoter by binding immediately upstream of start sites, and can unwind DNA. Related references: PMID:22037172
- Bends promoter DNA into a U-turn shape via its HMG-box domains, facilitating RNA polymerase recruitment. Related references: PMID:1737790
- Required for mtDNA maintenance and copy number regulation; plays a role in organizing and compacting mitochondrial nucleoids. Related references: PMID:19304746, PMID:22841477, PMID:22037171
- Localizes to the mitochondrial matrix and mitochondrial nucleoid.
- Post-translationally modified by phosphorylation; classified as a phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 100-200 of human TFAM; for applications requiring native conformation (e.g., IF/ICC), verify that the epitope is accessible.
- For Western blotting, the predicted molecular weight is approximately 29 kDa, but validate in your sample system.
- When using for IHC-P, optimize antigen retrieval and antibody dilution; cross-reactivity with mouse and rat tissues should be confirmed.
- As a polyclonal antibody, lot-to-lot consistency should be validated for critical assays; consider using appropriate controls.
