Biological Background: TET1 Function and Localization
- TET1 (Ten-eleven translocation 1) is a methylcytosine dioxygenase, also known as CXXC-type zinc finger protein 6 (CXXC6) or leukemia-associated protein with a CXXC domain (LCX).
- Catalyzes the sequential oxidation of 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC), 5-formylcytosine (5fC), and 5-carboxylcytosine (5caC), driving active DNA demethylation. Related references: PMID:19372391 PMID:21496894 PMID:21778364
- Recruits histone-modifying protein complexes to alter histone marks and chromatin accessibility, contributing to both gene activation and repression. Related references: PMID:33833093
- Plays essential roles in stem cell pluripotency, embryonic development, and differentiation; cooperates with QSER1 to protect transcriptional programs by inhibiting DNMT3A/3B-mediated de novo methylation. Related references: PMID:33833093
- Alternative splicing generates isoforms with distinct activities: isoform 1 broadly regulates synaptic transmission and memory, while isoform 2 localizes to replication sites in heterochromatin, preferentially demethylating LINE1 elements. (By similarity)
- Predominantly localizes to the nucleus and associates with chromosomes.
- Expressed in fetal heart, lung, and brain; adult skeletal muscle, thymus, and ovary; strongly upregulated in glioblastoma. Related references: PMID:25284789
- Contains a CXXC zinc-finger DNA-binding domain; requires iron(II) as a cofactor; subject to phosphorylation, glycosylation, and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 1800–1900 of human TET1; confirm epitope conservation if detecting mouse or rat TET1, as sequence divergence may affect cross-reactivity.
- For western blotting: use a low-percentage polyacrylamide gel (e.g., 6–8%) and extended transfer conditions for the ~235 kDa protein. Include a positive control lysate from human, mouse, or rat cells known to express TET1 (e.g., embryonic stem cells, glioblastoma lines).
- For immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P): optimize fixation/permeabilization protocols and confirm the expected nuclear/chromosomal staining pattern.
- Validate the antibody in your specific application and sample type; polyclonal antibodies can exhibit lot-to-lot variability, so routine quality-control testing is advised.
