  • TERF2IP (also known as RAP1, DRIP5) is a component of the shelterin complex, which protects telomere ends and regulates telomere length.
  • It negatively regulates telomere recombination by repressing homology-directed repair (HDR), thereby maintaining telomere length homeostasis; it is recruited to telomeric TTAGGG repeats via interaction with TERF2.
  • Beyond telomeres, TERF2IP acts as a transcription regulator at extratelomeric TTAGGG sites, modulating gene expression through association with TERF2 or other factors.
  • In the cytoplasm, TERF2IP associates with the IKK complex and promotes NF-κB signaling by enhancing IKK-mediated phosphorylation of RELA/p65, leading to activation of NF-κB target genes.
  • Subcellular localization includes the nucleus (telomeres and extratelomeric sites), cytoplasm, and chromosome territories.
