Biological Background: Tenascin C Function and Localization

  • Tenascin C (TN-C) is a large extracellular matrix glycoprotein belonging to the tenascin family, also known as cytotactin, hexabrachion, neuronectin, and glioma-associated extracellular matrix antigen. It is encoded by the TNC gene (Gene ID 3371).
  • In the central nervous system, TN-C guides migrating neurons and axons during development, promotes neurite outgrowth from cortical neurons, and participates in synaptic plasticity and neuronal regeneration.
  • TN-C serves as a ligand for several integrins, including alpha-8/beta-1, alpha-9/beta-1, alpha-V/beta-3, and alpha-V/beta-6, mediating cell–matrix interactions.
  • In tumors, TN-C stimulates angiogenesis by promoting endothelial cell elongation, migration, and sprouting. Related references: PMID:19884327
  • Subcellular localization: Secreted into the extracellular space and extracellular matrix.
  • Tissue specificity: Detected in fibroblasts at the protein level.
  • Additional features: Contains EGF-like domains, fibronectin type-III repeats, and a C-terminal fibrinogen domain; subject to alternative splicing and phosphorylation; associated with deafness and disease variants.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (amino acids 1990–2201) of human Tenascin C; for detection of endogenous full-length protein (approx. 241 kDa), consider using reducing SDS-PAGE and validate in relevant sample systems.
  • Polyclonal antibodies may recognize multiple isoforms or degraded fragments; confirm specificity using appropriate controls and, if necessary, isoform-specific reagents.
  • For immunohistochemistry (IHC-P) on paraffin-embedded sections, antigen retrieval may be required due to the extracellular matrix localization of TN-C.
  • For immunofluorescence (IF/ICC), include permeabilization steps and optimize fixation conditions, as secreted matrix proteins can be challenging to detect.
  • Cross-reactivity with mouse and rat has been reported; verify performance in your specific context, as epitope conservation may vary.
