- Tyrosine-protein kinase Tec (encoded by the TEC gene; also known as PSCTK4) is a non-receptor tyrosine kinase belonging to the Tec family of kinases, characterized by a pleckstrin homology (PH) domain, Tec homology (TH) domain, Src homology 2 and 3 (SH2 and SH3) domains, a kinase domain, and a zinc-finger motif.
- Tec functions as a signal transducer in multiple receptor-mediated pathways, regulating the actin cytoskeleton, and plays a critical role in the adaptive immune response by participating in T-cell receptor (TCR) and B-cell receptor (BCR) signaling.
- In T-cells, Tec acts redundantly with ITK to regulate development, differentiation, and function; it is required for TCR-dependent IL2 gene induction and contributes to CD28 co-stimulatory signaling by phosphorylating the adapter protein DOK1.
- In B-cells, Tec redundantly supports BTK in BCR-mediated signaling for development and activation, notably by phosphorylating STAP1, a BCR-signaling protein.
- Beyond lymphocytes, Tec is involved in mast cell cytokine production, myeloid cell growth and differentiation via CSF3 (G-CSF) signaling, and platelet activation downstream of integrin and G protein-coupled receptor pathways.
