  • TDP2 is a DNA repair hydrolase that specifically removes 5'-tyrosyl adducts from DNA, resolving topoisomerase 2 (TOP2)-DNA dead-end complexes and generating clean DNA double-strand breaks with 5'-phosphate termini suitable for ligation, thereby protecting neuronal gene transcription from abortive TOP2 activity.   • Functions in the cellular stress response by regulating ribosome biogenesis and participates in non-canonical TGF-beta signaling as an adaptor, promoting TRAF6-TAK1 complex ubiquitination and activation.

  • Also possesses weak 3'-tyrosyl DNA phosphodiesterase activity; represents the major, if not sole, 5'-tyrosyl DNA phosphodiesterase in cells, and hydrolyzes 5'-phosphoglycolates at radiation-induced DNA break ends.

  • During picornavirus infection, acts as a 5'-tyrosyl RNA phosphodiesterase (VPg unlinkase) to cleave the viral VPg protein from genomic RNA, a step critical for viral replication.
