Biological Background: TDP-43/TARDBP Function and Localization

  • TDP-43 is an RNA-binding protein with two RNA recognition motifs (RRM1 and RRM2) that preferentially binds GU-repeat sequences in long introns and 3'UTRs of mRNAs. In turn, it regulates the splicing of many non-coding and protein-coding RNAs, including those involved in neuronal survival and neurodegenerative diseases. Related references: PMID:23519609 PMID:24240615 PMID:24464995 PMID:21358640 PMID:29438978
  • Plays a role in maintaining mitochondrial homeostasis by regulating the processing of mitochondrial transcripts. Related references: PMID:28794432
  • Regulates mRNA stability by recruiting CNOT7/CAF1 deadenylase to mRNA 3'UTR, leading to poly(A) tail deadenylation and thus shortening. Related references: PMID:30520513
  • In response to oxidative insult, associates with stalled ribosomes localized to stress granules (SGs) and contributes to cell survival. Related references: PMID:19765185 PMID:23398327
  • Participates in normal skeletal muscle formation and regeneration, forming cytoplasmic myo-granules and binding mRNAs that encode sarcomeric proteins. Related references: PMID:30464263
  • Contributes to the maintenance of circadian clock periodicity via stabilization of CRY1 and CRY2 proteins in an FBXL3-dependent manner. Related references: PMID:27123980
  • Negatively regulates the expression of CDK6 and modulates HDAC6, ATG7, and VCP expression in a PPIA/CYPA-dependent manner. Related references: PMID:19760257 PMID:25678563
  • Localizes to the nucleus, cytoplasm, stress granules, and mitochondria. Ubiquitously expressed, with particularly high levels in pancreas, placenta, lung, genital tract, and spleen.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-260) of human TDP-43, which includes the nuclear localization signal and a portion of RRM1. Consider potential effects on epitope accessibility in native protein interaction assays like IP or RIP.
  • For Western blot (WB), a major band around 45 kDa is expected. However, TDP-43 may form higher-molecular-weight complexes or aggregates; optimize reducing conditions and sample preparation accordingly.
  • Validate antibody performance in your specific experimental system using appropriate positive controls for human, mouse, or rat samples, as cross-reactivity has been reported.
  • In immunohistochemistry (IHC-P), antigen retrieval may be necessary. Expect both nuclear and cytoplasmic staining based on the protein's dual localization patterns.
  • For RIP assays, include RNase-treated controls to confirm RNA-dependent interactions and ensure specificity.
