Biological Background: TDP-43/TARDBP Function and Localization
- TAR DNA-binding protein 43 (TDP-43) is encoded by the TARDBP gene and belongs to the hnRNP family of RNA-binding proteins.
- It preferentially binds GU-rich RNA sequences via its two RRM domains, regulating pre-mRNA splicing, mRNA stability, and transport. Related references: PMID:23519609, PMID:24240615, PMID:24464995
- TDP-43 is critical for the alternative splicing of transcripts involved in neuronal survival and neurodegeneration. Related references: PMID:21358640, PMID:29438978
- Mediates mRNA deadenylation and decay by recruiting the CNOT7 deadenylase to 3'UTRs. Related references: PMID:30520513
- Under oxidative stress, translocates to cytoplasmic stress granules and associates with stalled ribosomes to promote cell survival. Related references: PMID:19765185, PMID:23398327
- Regulates circadian clock periodicity by stabilizing CRY1/CRY2 proteins in an FBXL3-dependent manner. Related references: PMID:27123980
- Localizes predominantly to the nucleus but also to the cytoplasm, stress granules, and mitochondria.
- Ubiquitously expressed with enrichment in pancreas, placenta, lung, genital tract, and spleen.
Experimental Guidance and Technical Tips
- For Western blotting, empirically determine the optimal primary antibody dilution; expect a specific band near 45 kDa corresponding to TDP-43.
- When performing IHC-P or IF/ICC, optimize antigen retrieval and permeabilization protocols due to the protein's nuclear and cytoplasmic localization.
- Validate cross-reactivity in mouse and rat tissues if these species are used, as indicated by the supplier's cross-reactivity data.
- The immunogen (N-terminal 1-100 amino acids) may recognize both full-length and N-terminal isoforms; consider this in isoform-specific experiments.
- ELISA can be employed for quantification; ensure proper coating concentrations and blocking conditions are established.
