Biological Background: TDP-43 Function and Localization
- TAR DNA-binding protein 43 (TDP-43) is encoded by the TARDBP gene and is also known as ALS10. It belongs to the hnRNP family and contains two RNA recognition motifs (RRMs) and a glycine-rich C-terminal domain.
- Functions as an RNA-binding protein involved in multiple steps of RNA biogenesis and processing (PubMed:23519609). Related references: PMID:23519609
- Preferentially binds GU-repeats via its RRMs, localizing to long introns and 3'UTRs of mRNAs, thereby regulating splicing of many RNAs, including those critical for neuronal survival and neurodegeneration (PubMed:23519609, PubMed:24240615, PubMed:24464995, PubMed:21358640, PubMed:29438978). Experimental Guidance and Technical Tips
- The immunogen targets the C-terminal region (aa 300–400) of human TDP-43, which may recognize both full-length and cleaved forms; consider this when interpreting band patterns in Western blot.
- For Western blot, a predominant band near 45 kDa is expected; use appropriate positive controls such as HeLa or brain tissue lysates and validate in your sample system.
- Cross-reactivity with mouse and rat has been documented; confirm performance in your experimental settings, especially for immunohistochemistry where fixation and antigen retrieval may affect staining.
- In immunohistochemistry (IHC), optimize antigen retrieval (e.g., citrate buffer pH 6.0 or EDTA pH 9.0) and detection conditions for each tissue type.
- For immunoprecipitation (IP) and chromatin immunoprecipitation (ChIP), verify antibody suitability for native protein binding; note that TDP-43 is primarily an RNA-binding protein, and DNA interactions may require cross-linking optimization.
- This antibody has been used in ELISA; however, individual assay conditions (e.g., coating buffer, blocking agent) should be optimized to achieve desired sensitivity.
