Biological Background: TDGF1/CRIPTO Function and Localization

  • Protein Cripto (Teratocarcinoma-derived growth factor 1), encoded by the TDGF1/CRIPTO gene, is a GPI-anchored cell membrane protein belonging to the EGF-CFC family. It is also known as CR, CR-1, CRGF, or CRIPTO.
  • Acts as a coreceptor for Nodal, a member of the TGF-beta superfamily. Cell-associated CRIPTO facilitates Nodal signaling in cis, while shedding by TMEM8A generates a soluble form that can function in trans on neighboring cells.

  • Plays a role in early embryonic development, particularly in the determination of epiblastic cells that give rise to the mesoderm.

  • Subcellular localization includes the cell membrane and secretion into the extracellular space, consistent with its GPI anchor and shedding.
  • Preferentially expressed in gastric and colorectal carcinomas compared to normal tissues, and also detected in breast and lung cancers.
  • Features an EGF-like domain, multiple disulfide bonds, and glycosylation sites. It is synthesized with a signal peptide and anchored to the membrane via a GPI moiety.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 31-150 of human TDGF1, which excludes the signal peptide (residues 1-30) and includes the EGF-like domain. This design likely enhances specificity for mature and processed forms of the protein.
  • For Western blot (WB), a band around 21 kDa may be observed, but post-translational modifications such as glycosylation and GPI-anchor addition can result in higher molecular weight species. Validate detection in relevant cancer cell lines (e.g., gastric, colorectal, breast, lung).
  • In immunohistochemistry (IHC-P), membrane and/or cytoplasmic staining is expected. Ensure proper antigen retrieval and consider using cancer tissue sections with known CRIPTO overexpression.
  • The antibody shows cross-reactivity with mouse and rat TDGF1, making it suitable for studies in rodent models; however, validate species-specific reactivity in your experimental setup.
