Biological Background: Translationally-controlled tumor protein (TCTP/TPT1) Function and Localization

  • Translationally-controlled tumor protein (TCTP), also known as fortilin, histamine-releasing factor (HRF), and p23, is encoded by the TPT1 gene.
  • TCTP is involved in calcium binding and microtubule stabilization, supporting cellular structural integrity and signaling (PubMed:12167714, PubMed:15162379, PubMed:15958728). Related references: PMID:12167714 PMID:15162379 PMID:15958728
  • It acts as a negative regulator of TSC22D1-mediated apoptosis by interacting with and destabilizing the TSC22D1 protein (PubMed:18325344). Related references: PMID:18325344
  • The protein is predominantly localized in the cytoplasm.
  • TCTP is expressed in a wide range of healthy and tumoral cells including erythrocytes, hepatocytes, macrophages, platelets, keratinocytes, erythroleukemia cells, gliomas, melanomas, hepatoblastomas, and lymphomas, but is notably absent in kidney and renal cell carcinoma. It is also expressed in placenta and prostate.
  • Post-translational modifications include phosphorylation; alternative splicing produces multiple isoforms. The protein has a known 3D structure and is part of the reference proteome.
