Biological Background: TCTP/TPT1 Function and Localization
- TCTP/TPT1, encoded by the TPT1 gene, is also known as Translationally-controlled tumor protein, Fortilin, Histamine-releasing factor (HRF), and p23.
- It plays a role in calcium binding and microtubule stabilization, contributing to cytoskeletal dynamics and cellular integrity. Related references: PMID:12167714, PMID:15162379, PMID:15958728
- TCTP acts as a negative regulator of apoptosis by interacting with and destabilizing TSC22D1 protein, thereby inhibiting TSC22D1-mediated cell death. Related references: PMID:18325344
- The protein is predominantly localized in the cytoplasm.
- It is widely expressed in healthy and tumoral cells, including erythrocytes, hepatocytes, macrophages, and various cancer types, but absent in kidney and renal cell carcinoma. Expressed in placenta and prostate.
- TCTP is subject to phosphorylation and alternative splicing, as indicated by proteomics and 3D-structure studies.
- Its molecular weight is approximately 20 kDa (calculated 19.6 kDa), and it belongs to the TPT1 family.
Experimental Guidance and Technical Tips
- The immunogen encompasses the full-length human TCTP sequence (amino acids 1-172), which may facilitate recognition of native and recombinant TCTP proteins across species.
- For Western blot (WB) experiments, consider using denaturing and reducing conditions, as the predicted/observed molecular weight is ~20 kDa.
- In ELISA applications, the antibody may be suitable for both direct and sandwich formats; cross-reactivity with mouse and rat TCTP should be validated in the relevant sample system.
- Given TCTP's ubiquitous expression, appropriate positive and negative control lysates (e.g., placenta vs. kidney) can help ensure specificity.
