Biological Background: TCTP/TPT1 Function and Localization

  • TCTP (Translationally-controlled tumor protein), also known as Fortilin, histamine-releasing factor (HRF), and p23, is encoded by the TPT1 gene.

  • TCTP acts as a negative regulator of TSC22D1-mediated apoptosis by interacting with and destabilizing TSC22D1 protein. Related references: PMID:18325344
  • The protein localizes to the cytoplasm.
  • TCTP is widely expressed in healthy and tumoral cells, including erythrocytes, hepatocytes, macrophages, platelets, keratinocytes, and various tumors; however, it is not detected in kidney and renal cell carcinoma (RCC). It is also expressed in placenta and prostate.
  • Post-translational modifications include phosphorylation, consistent with its classification as a phosphoprotein.
  • Alternative splicing produces multiple isoforms of TCTP.
