Biological Background: T-cell Leukemia Translocation-altered Gene Protein (TCTA) Function and Localization
- TCTA (T-cell leukemia translocation-altered gene protein) is a proto-oncogene originally identified through chromosomal rearrangement in T-cell leukemia. The protein is also known as T-cell leukemia translocation-associated gene protein.
- It is a small transmembrane protein with a calculated molecular weight of ~11 kDa, consisting of 103 amino acids.
- TCTA may play a role in cellular fusion during osteoclastogenesis, contributing to bone resorption and remodeling.
- The protein localizes to the membrane and possesses a single transmembrane helix, classifying it as an integral membrane protein.
- TCTA is expressed ubiquitously, with the highest levels in the kidney. At the protein level, it is detected in monocytes, osteoclasts, macrophages, synoviocytes, and synovial lining cells.
- Post-translational modifications include N-linked glycosylation and acetylation, which may regulate its function or stability.
Experimental Guidance and Technical Tips
- For Western blotting (WB), the predicted molecular weight is 11 kDa. Consider using high-concentration gels (e.g., 15%) and low-molecular-weight markers, along with careful transfer conditions to prevent loss of small proteins.
- In immunofluorescence/ICC, TCTA is a membrane protein. Employ appropriate permeabilization and blocking steps to ensure antibody access. Validate the staining pattern in known expressing cell types such as osteoclasts or macrophages.
- For ELISA, the antibody can be used as a detection reagent; however, optimal coating and detection conditions should be empirically determined for the specific assay format.
