Biological Background: TCP1 beta/CCT2 Function and Localization
- TCP1 beta (CCT2, CCT-beta) is the beta subunit of the chaperonin-containing T-complex (TRiC), a group II chaperonin essential for protein folding in the cytoplasm.
- As part of TRiC, TCP1 beta participates in the ATP-dependent folding of newly synthesized actin and tubulin, critical for cytoskeletal integrity and cell division. Related references: PMID:25467444 PMID:36493755 PMID:35449234
- TRiC also folds WRAP53/TCAB1, a component required for telomere maintenance, highlighting its role in genome stability. Related references: PMID:25467444
- Additionally, TRiC contributes to the assembly of the BBSome, a multiprotein complex that regulates vesicular trafficking to the primary cilium, important for ciliary signaling. Related references: PMID:20080638
- TCP1 beta localizes to the cytoplasm, consistent with its function as a molecular chaperone.
- The protein features an ATP-binding domain and undergoes post-translational modifications such as acetylation and phosphorylation, which may regulate its chaperonin activity.
Experimental Guidance and Technical Tips
- The immunogen spans the C-terminal region (aa 206-535), which may enable recognition of both native and denatured TCP1 beta; however, cross-reactivity with other CCT subunits should be tested.
- For Western blot, a band corresponding to ~57 kDa is expected in human and mouse cell lysates. Validate with appropriate controls.
- For immunofluorescence, cytoplasmic staining should be observed; consider co-staining with organelle markers.
- In immunoprecipitation, this polyclonal antibody may efficiently capture the TCP1 beta subunit and associated TRiC complex components; optimization of lysis and wash conditions is recommended.
- For ELISA, the antibody can be used in both direct and sandwich formats; pairing with a monoclonal anti-TCP1 beta antibody may enhance sensitivity.
