Biological Background: TCL1A Function and Localization
- TCL1A (T-cell leukemia/lymphoma protein 1A) is a proto-oncogene initially identified through chromosomal rearrangements in T-cell leukemias and lymphomas. It is also known as oncogene TCL1 or protein p14 TCL1.
- The protein functions as an AKT co-activator, enhancing the phosphorylation and activation of AKT1, AKT2, and AKT3, and promoting the nuclear translocation of AKT1.
- TCL1A contributes to enhanced cell proliferation and survival by stabilizing the mitochondrial membrane potential.
- Subcellularly, TCL1A is found in the cytoplasm, nucleus, microsomes, and endoplasmic reticulum.
- Expression is predominantly restricted to immature thymocytes and activated peripheral lymphocytes, with preferential expression early in T- and B-lymphocyte differentiation.
- The predicted molecular weight of TCL1A is approximately 13.5 kDa, consistent with the observed band at 13 kDa.
Experimental Guidance and Technical Tips
- The immunogen consists of the full-length TCL1A protein (amino acids 1-114), which may allow recognition of native and denatured forms; consider this when selecting assay conditions.
- For Western blot, the protein migrates at approximately 13 kDa; ensure adequate separation and transfer of low-molecular-weight proteins.
- ELISA can be used to detect TCL1A in cell lysates or recombinant proteins; cross-reactivity with other TCL family members should be assessed due to the polyclonal nature of the antibody.
- Validate antibody performance in the relevant biological matrices, such as human lymphoid cell lines or primary lymphocytes, to confirm specificity and sensitivity.
