- TCF7L1, also known as TCF-3 or HMG box transcription factor 3, is a member of the TCF/LEF family of high-mobility group (HMG) box transcription factors that mediate canonical Wnt signaling.
- In the absence of Wnt/β-catenin signaling, TCF7L1 acts as a transcriptional repressor by binding to Wnt-responsive DNA elements; upon pathway activation and β-catenin (CTNNB1) accumulation, it switches to an activator, promoting target gene expression.
- TCF7L1 is essential for terminal differentiation of epidermal cells, formation of keratohyalin granules, and establishment of the skin barrier function, as demonstrated by similarity studies.
