Biological Background: TCF4/TCF7L2 Function and Localization
- Transcription factor 7-like 2 (TCF7L2), also known as TCF-4, TCF4, or hTCF-4, is a member of the HMG box transcription factor family.
- Functions as a key effector of the Wnt signaling pathway: acts as a transcriptional repressor in the absence of β-catenin (CTNNB1) and as an activator upon β-catenin binding.
- Binds the Tcf motif 5'-CCTTTGATC-3' in target gene promoters; regulates expression of MYC and other Wnt-responsive genes.
- Necessary for maintenance of the epithelial stem-cell compartment in the small intestine; expression of dominant-negative mutants induces G1 cell-cycle arrest.
- Primarily localizes to the nucleus and PML nuclear bodies.
- Detected in small intestinal epithelium (highest at crypt top), colon epithelium, colon cancer, and mammary gland epithelium and carcinomas.
- Post-translationally modified by phosphorylation and ubiquitination (UBL conjugation); contains isopeptide bonds.

Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against a recombinant fragment corresponding to amino acids 124–188 of human TCF4/TCF7L2 (isoform NP_001354872.1). Due to alternative splicing, this epitope may be present in some but not all isoforms; verify expression in your model system.
- For Western blotting, use nuclear-enriched lysates from human or mouse intestinal or colon epithelial cells/tissues to enrich for TCF4/TCF7L2. Expect a band around 68 kDa; multiple bands may appear due to isoform diversity and post-translational modifications.
- For ELISA, consider using recombinant TCF4/TCF7L2 fragment as coating antigen and validate with known positive and negative control sera or purified antibodies.
- Optimize antibody dilution and incubation conditions for each application; include appropriate controls (e.g., siRNA knockdown or knockout cell lysates) to confirm specificity.
- Cross-reactivity with human and mouse has been indicated; reactivity with other species should be tested if needed.
