Biological Background: TCF4/TCF7L2 Function and Localization
- Transcription factor 7-like 2, also known as T-cell factor 4 (TCF-4), is a member of the TCF/LEF family of DNA-binding transcription factors.
- Functions as a key effector in the canonical Wnt/β-catenin signaling pathway, regulating gene expression by binding to Tcf motifs (5'-CCTTTGATC-3') in target gene promoters.
- Acts as a transcriptional repressor in the absence of β-catenin (CTNNB1) and as an activator upon β-catenin binding, switching target gene expression.
- Modulates MYC expression by binding to its promoter, and regulates cell-cycle progression; dominant-negative mutants cause G1 arrest.
- Essential for maintaining the epithelial stem-cell compartment of the small intestine, highlighting its role in tissue homeostasis.
- Predominantly localized in the nucleus and PML bodies, consistent with its role in transcriptional regulation.
- Detected in the epithelium of the small intestine (highest at crypt tops), colon, and mammary gland, with elevated expression in colon and mammary carcinomas.
- Subject to phosphorylation and ubiquitin-like conjugation (ISGylation), influencing activity and stability (Keywords: Phosphoprotein, Ubl conjugation, Isopeptide bond).

- The immunogen corresponds to the N-terminal region (a.a. 1-100), which may allow detection of multiple TCF7L2 isoforms if this region is conserved.
- For Western blot (WB), a band around 68 kDa is expected; validate in human cell lines known to express TCF7L2, such as colorectal cancer cells.
- For immunohistochemistry (IHC-P), antigen retrieval and antibody dilution should be optimized for formalin-fixed paraffin-embedded (FFPE) human tissue sections, particularly intestinal or colon samples where expression is reported.
- For ELISA, consider using recombinant TCF7L2 protein or peptide fragments covering the immunogen sequence for standard curve generation.
- Since TCF7L2 activity is context-dependent (Wnt pathway), co-treatment with Wnt agonists or β-catenin stabilizers may be considered to study activation states.
