Biological Background: TCF4 Function and Localization
- Transcription factor 4 (TCF4), also known as immunoglobulin transcription factor 2 (ITF-2) and SEF-2, is a member of the class B basic helix-loop-helix (bHLH) protein family (bHLHb19). It plays a critical role in transcriptional regulation and neuronal differentiation.
- Binds to the immunoglobulin enhancer Mu-E5/KE5-motif and activates transcription by recognizing E-box sequences (5'-CANNTG-3'), with a preference for either 5'-ACANNTGT-3' or 5'-CCANNTGG-3'. It also binds to the E-box in the somatostatin receptor 2 initiator element (SSTR2-INR) (By similarity).
- Involved in the initiation of neuronal differentiation, consistent with its expression in developing embryonic brain and adult brain, heart, placenta, and skeletal muscle.
- Localizes to the nucleus, where it exerts its DNA-binding and transcription regulation functions.
- Post-translational modifications include phosphorylation, and the protein undergoes alternative splicing, generating multiple isoforms.
- Associated with various diseases, including corneal dystrophy, epilepsy, intellectual disability, and primary microcephaly, as indicated by disease variant annotations.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 1–210 of human TCF4 (N-terminal region). For Western blot, the predicted molecular weight is ~71 kDa; however, alternative splicing or post-translational modifications may result in additional bands.
- IHC-P and IF/ICC: Nuclear staining is expected. Consider adequate fixation and permeabilization protocols for optimal antigen detection.
- Due to the polyclonal nature and bHLH domain homology, potential cross-reactivity with other bHLH proteins should be evaluated. Use of knockout or knockdown controls is recommended.
- ELISA: Suitable for capture or detection of TCF4 protein. Validate the assay conditions in the relevant sample matrix.
