- Encodes transcription factor E2-alpha, a class B basic helix-loop-helix (bHLH) protein also known as TCF3, bHLHb21, or immunoglobulin enhancer-binding factor E12/E47.
- Functions as a transcriptional regulator that initiates neuronal differentiation and promotes mesenchymal to epithelial transition by forming heterodimers with tissue-specific bHLH proteins.
- Binds to E-box DNA motifs (5'-CANNTG-3') and regulates tissue-specific gene expression during embryogenesis, including in muscle and early B-cell differentiation.
- Recognizes the kappa-E2 site in the immunoglobulin gene enhancer, implicating it in B-cell development. Related references: PMID:2493990
- Interacts with the insulin gene transcription control region, contributing to pancreatic cell regulation.
- Enhances ATOH7 binding to the consensus sequence 5'-CAGGTG-3', positively modulating transcriptional activity.
