Biological Background: TCF1/TCF7 Function and Localization

  • Transcription factor 7 (TCF7), also known as TCF-1, is a member of the TCF/LEF family of high mobility group (HMG) box DNA-binding proteins.
  • TCF7 acts as a transcriptional activator essential for T-cell lymphocyte differentiation and survival of CD4+CD8+ immature thymocytes; isoforms lacking the N-terminal CTNNB1 binding domain cannot support this function.
  • The protein binds the T-lymphocyte-specific enhancer element (5'-WWCAAAG-3') in the CD3E gene promoter and represses T-cell receptor gamma gene expression in alpha-beta T-cell lineages (By similarity).
  • TCF7 is required for development of natural killer receptor-positive lymphoid tissue inducer T-cells (By similarity).
  • Transcriptional activity is repressed by TLE family members (TLE1-4) and may act as a feedback repressor of CTNNB1 and TCF7L2 target genes.
  • Subcellular localization is predominantly nuclear, consistent with its role in transcription regulation.
  • TCF7 is expressed predominantly in T-cells, with additional detection in proliferating intestinal epithelial cells and basal epithelial cells of mammary gland epithelium.
  • Keywords highlight its involvement in the Wnt signaling pathway and dual activator/repressor functions.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 50–150 of human TCF7 (NP_003193.2). Based on this sequence, the antibody may recognize multiple isoforms; consider verifying the detected band pattern in your specific sample type.
  • For Western blot, TCF7 has a predicted molecular weight of approximately 42 kDa, but post-translational modifications or isoform variants may alter migration; nuclear extracts often improve detection sensitivity.
  • The antibody is validated for WB and ELISA. For WB, a starting dilution of 1:500 to 1:1000 is commonly used for polyclonal antibodies; optimize for your system.
  • Cross-reactivity with mouse and rat is indicated, making it suitable for rodent models; however, confirm species-specific reactivity in pilot experiments.
  • Since TCF7 is a transcription factor predominantly expressed in T-cells, consider using T-cell-rich samples (e.g., thymus, spleen) as positive controls.
