Biological Background: TCF1/TCF7 Function and Localization

  • TCF1/TCF7 (Transcription factor 7), also known as T-cell factor 1 (TCF-1), is a DNA-binding transcriptional activator crucial for T-cell lymphocyte differentiation and the survival of CD4+CD8+ immature thymocytes.
  • The protein binds to the T-lymphocyte-specific enhancer element (5'-WWCAAAG-3') in the CD3E gene promoter, and represses expression of the T-cell receptor gamma gene in alpha-beta T-cell lineages.
  • Isoforms lacking the N-terminal CTNNB1 (β-catenin) binding domain are unable to support immature thymocyte survival, highlighting the role of Wnt/β-catenin signaling in T-cell development.
  • TCF7 acts as a feedback repressor of CTNNB1 and TCF7L2 target genes, and its transactivation is inhibited by TLE family co-repressors (TLE1, TLE2, TLE3, TLE4).
  • Required for the development of natural killer receptor-positive lymphoid tissue inducer T-cells, indicating roles in innate lymphoid cell biology.

  • The immunogen is a synthetic peptide spanning amino acids 50–150 of human TCF1/TCF7 (NP_003193.2). Given the location, the antibody may detect multiple isoforms; verify epitope availability in your experimental context.
  • For western blotting, expect a band around 42 kDa corresponding to the full-length protein, but additional bands from splice variants or post-translational modifications may be present. Use T-cell lysates (e.g., Jurkat) as a positive control, and consider mouse/rat cross-reactivity for model organism studies.
  • In ELISA applications, include peptide or recombinant TCF1/TCF7 as a positive control, and perform titration to determine optimal working concentration.
