Biological Background: TCF12 Function and Localization
- TCF12 (Transcription factor 12) is a member of the basic helix-loop-helix (bHLH) family of transcription factors, also known as HEB, HTF4, and bHLHb20.
- It functions as a transcriptional regulator that activates transcription by binding to the E-box motif (5'-CANNTG-3') on target gene promoters (By similarity).
- Involved in the initiation of neuronal differentiation, playing a critical role in the development of the gonadotropin-releasing hormone (GnRH) axis. Related references: PMID:32620954
- Localizes predominantly to the nucleus, consistent with its role in transcriptional regulation.
- TCF12 is associated with diseases such as craniosynostosis, hypogonadotropic hypogonadism, and Kallmann syndrome, and is marked by keywords including alternative splicing, phosphoprotein, and Ubl conjugation.
- Expression is detected in several tissues including skeletal muscle, thymus, and B-cell lines.
Experimental Guidance and Technical Tips
- For Western blot, consider using lysates from human, mouse, or rat tissues such as skeletal muscle or thymus, where TCF12 is expressed. The predicted molecular weight is 73 kDa; verify appropriate band migration.
- In ELISA, consider using the recombinant immunogen as a positive control to assess antibody binding.
- This polyclonal antibody recognizes multiple epitopes; for optimal results, perform titration experiments to determine the best working dilution for your specific assay conditions.
- Note that TCF12 is a nuclear protein; appropriate nuclear extraction or permeabilization may enhance detection in cell-based assays.
