Biological Background: Elongin-B (TCEB2) Function and Localization

  • Elongin-B (TCEB2) is the 18 kDa subunit of the transcription elongation factor SIII (elongin), which enhances RNA polymerase II elongation past template-encoded arrest sites. The transcriptional activity of subunit A is strongly enhanced by binding to the elongin BC complex. Related references: PMID:7638163
  • Core component of multiple cullin-2 and cullin-5-RING E3 ubiquitin-protein ligase complexes (ECS complexes), mediating ubiquitination of target proteins. It links substrate recruitment subunits (like VHL, SOCS box proteins) to Cullin/RBX1 modules. Related references: PMID:10205047, PMID:12004076, PMID:12050673
  • Involved in the DesCEND pathway, where various ECS complexes recognize C-terminal degrons to ubiquitinate and degrade target proteins. Related references: PMID:26138980, PMID:29775578, PMID:29779948
  • Participates in specific ubiquitination events: with ECS(ASB9) it ubiquitinates CKB; with ECS(CBC(VHL)) it polyubiquitinates monoubiquitinated POLR2A; with ECS(ASB7) it mediates degradation of SUV39H1, regulating H3K9me3; with ECS(LRRC58) it degrades CDO1. Related references: PMID:33268465, PMID:19920177, PMID:40440427, PMID:40963025
  • Subcellularly localized to the nucleus, consistent with its roles in transcription and ubiquitination.
  • Subject to post-translational modifications including acetylation and phosphorylation, as indicated by proteomics studies.
  • Hijacked by HIV-1 Vif to form an ECS complex that ubiquitinates and degrades APOBEC3F/G, facilitating viral immune evasion. Related references: PMID:18562529, PMID:20532212, PMID:22190037

Experimental Guidance and Technical Tips

  • The immunogen (full-length amino acids 1-118) suggests detection of native and denatured TCEB2; suitable for WB, IP, and ELISA. For IHC-P, optimize antigen retrieval.
  • As a polyclonal antibody, confirm reactivity in the relevant human sample system and consider potential batch-to-batch consistency checks.
  • For co-immunoprecipitation or ubiquitination assays, validate antibody specificity under the experimental conditions used.
  • The nuclear localization of TCEB2 may require appropriate fractionation or permeabilization steps in IF or IHC.
