Biological Background: Elongin-C (TCEB1) Function and Localization
- Elongin-C (TCEB1), also known as SIII p15, is the 15 kDa subunit of the general transcription elongation factor SIII, which increases RNA polymerase II transcription elongation past template-encoded arresting sites. Related references: PMID:7821821
- Elongin-C serves as a core component of multiple cullin-RING-based ECS (ElonginB/C-CUL2/5-SOCS-box protein) E3 ubiquitin-protein ligase complexes, mediating ubiquitination and degradation of diverse targets, including CKB, SUV39H1, CDO1, POLR2A, and MCM7. Related references: PMID:10205047, PMID:12004076, PMID:12050673, PMID:26138980, PMID:29775578, PMID:29779948, PMID:33268465, PMID:19920177, PMID:35512830, PMID:40440427, PMID:40963025
- During HIV-1 infection, Elongin-C is hijacked by viral Vif protein to form an ECS complex that ubiquitinates and degrades host antiviral factors APOBEC3F and APOBEC3G. Related references: PMID:18562529, PMID:20532212, PMID:22190037, PMID:37640699
- The protein is primarily localized to the nucleus, consistent with its roles in transcription and ubiquitination.
- TCEB1 is overexpressed in certain cancer cell lines, including prostate cancer PC-3 and breast cancer SK-BR-3, suggesting a potential role in oncogenesis.
- The gene undergoes alternative splicing and is involved in host-virus interactions and ubiquitin conjugation pathways.
Experimental Guidance and Technical Tips
- For Western blotting, a band at approximately 12 kDa is expected; validate with appropriate positive (e.g., HeLa, HEK293) and negative controls in your sample system.
- In immunohistochemistry (IHC-P), consider antigen retrieval methods and use cancer tissue sections (such as prostate or breast) as potential positive controls given the overexpression pattern.
- As a polyclonal antibody, it may detect multiple isoforms or degradation products; verify specificity using knockdown or knockout samples if available.
- The antibody cross-reacts with mouse and rat, enabling cross-species studies; optimize dilutions for each species and application.
- For ELISA, consider pairing with a matched detection antibody and validate using recombinant TCEB1 protein.
