Biological Background: TCEA1 Function and Localization
- TCEA1 (Gene ID: 6917) encodes transcription elongation factor A protein 1, also known as transcription elongation factor S-II protein 1 (TFIIS) and TFIIS.o.
- The protein localizes to the nucleus and plays a critical role in RNA polymerase II transcription elongation.
- It enables RNA polymerase II to read through template-encoded arrest sites by inducing cleavage of the nascent RNA transcript, thereby generating a new 3'-terminus for extension.
- TCEA1 belongs to the transcription elongation factor A family and contains a zinc-finger domain essential for its function.
- Post-translational modifications include acetylation, phosphorylation, isopeptide bond formation, and ubiquitination.
- The gene undergoes alternative splicing and has been reported to participate in chromosomal rearrangements.
Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic peptide spanning amino acids 50–150 of human TCEA1; this epitope is conserved in mouse and rat, consistent with observed cross-reactivity.
- For Western blotting, nuclear extracts or whole-cell lysates are recommended; a specific band at approximately 34 kDa is expected.
- In immunofluorescence/ICC, TCEA1 shows a predominantly nuclear localization pattern; appropriate fixation and permeabilization should be optimized.
- ELISA applications may be used for quantitative detection of TCEA1 in various sample types; titrate the antibody to determine optimal working concentration.
- Although the antibody is listed for human, mouse, and rat, performance in other species should be empirically validated.
