Biological Background: TBX3 Function and Localization

  • TBX3 is a T-box transcription factor that functions as a transcriptional repressor involved in numerous developmental processes. Related references: PMID:10468588 PMID:12000749
  • It binds specifically to the palindromic T site 5'-TTCACACCTAGGTGTGAA-3' DNA sequence or half-sites, regulating target gene expression. Related references: PMID:12000749
  • TBX3 plays a critical role in limb pattern formation and is required for mammary placode induction and maintenance of mammary buds. Related references: PMID:10468588
  • Together with TBX2, TBX3 is involved in branching morphogenesis of developing lungs and adult mammary glands, and maintains cell proliferation in embryonic lung mesenchyme, possibly downstream of SHH, BMP, and TGFβ signaling.
  • It modulates early inner ear development, acting independently and redundantly with TBX2 in different subregions.
  • TBX3 acts as a negative regulator of PML function in cellular senescence. Related references: PMID:22002537
  • The protein is localized to the nucleus and is widely expressed across tissues.
  • Multiple isoforms exist due to alternative splicing, including TBX3-ISO; the protein is subject to phosphorylation and is linked to disease variants.

Experimental Guidance and Technical Tips

  • When performing Western blotting, TBX3 is predicted at ~79 kDa; we recommend optimizing lysate preparation and antibody concentration to detect the target band.
  • For ELISA, the immunogen (aa 310-480) may enable detection of both native and recombinant TBX3; consider using recombinant protein standards for quantification.
  • Given the reported cross-reactivity with mouse, this antibody can be a candidate for mouse model studies; validation in your specific sample system is advised.
