Biological Background: TBX21 Function and Localization
- TBX21 is a lineage-defining transcription factor that drives Th1 cell development from naive precursors, simultaneously activating Th1-specific genetic programs while repressing Th2 and Th17 lineages. Related references: PMID:10761931
- It selectively activates genes critical for Th1 function, such as IFN-gamma and CXCR3, in part by recruiting chromatin remodeling complexes (KDM6B, SWI/SNF) and modifying epigenetic marks to establish a transcriptionally permissive state.
- TBX21 recruits the super elongation complex (SEC) containing Mediator and P-TEFb to super-enhancers of Th1-associated genes, enhancing their transcription. Related references: PMID:27292648
- It inhibits Th17 differentiation by blocking RUNX1-mediated RORC transactivation, and suppresses Th2 cytokines by repressing GATA3 and NFATC2.
- In an IFN-gamma-rich microenvironment, TBX21 acts as a repressor of type-I interferon responses to prevent aberrant amplification.
- Required for proper development of natural killer (NK) and mucosal-associated invariant T (MAIT) cells. Related references: PMID:33296702
- Localizes to the nucleus and shows T-cell-specific expression.
- Undergoes post-translational modifications including phosphorylation and ubiquitination (isopeptide bond formation).
Experimental Guidance and Technical Tips
- The immunogen spans the C-terminal region (aa 322-535) of human TBX21, which shares high homology across human, mouse, and rat, consistent with the reported multi-species cross-reactivity.
- For Western blotting, expect a band at approximately 58 kDa; validate using lysates from Th1-polarized T cells or tissues with elevated T-bet expression.
- ELISA protocols may require optimization of coating antigen concentration and pairing with a detection antibody; consider using recombinant T-bet protein as a standard.
- As a polyclonal antibody, lot-to-lot consistency should be verified for each application, and appropriate controls (e.g., TBX21 knockout or siRNA-treated samples) are recommended to confirm specificity.
