Biological Background: TBR1 Function and Localization

  • TBR1 (T-box brain protein 1, also known as TES-56) is a member of the T-box family of transcription factors.
  • Acts as a transcriptional repressor critical for cerebral cortex development, including neuronal migration and arealization (PubMed:25232744, PubMed:30250039). Related references: PMID:25232744 PMID:30250039
  • Represses FEZF2 to confine corticospinal tract origin to layer 5 neurons (By similarity).
  • Localizes to the nucleus.
  • Highly expressed in the brain.
  • Associated with neurodevelopmental disorders such as autism spectrum disorder and intellectual disability.
  • Undergoes alternative splicing and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide spanning amino acids 1-100 of human TBR1. The antibody may recognize full-length TBR1 and potentially N-terminal isoforms or cleavage products. Consider this when interpreting band patterns.
  • For Western blot (WB), TBR1 migrates at approximately 74 kDa. Use nuclear-enriched lysates from brain tissue or neuronal cell lines (e.g., SH-SY5Y) for optimal detection. Validate with a known positive control.
  • In immunofluorescence (IF-P), TBR1 localizes to the nucleus. Adequate permeabilization and fixation are essential. Test different conditions to achieve specific staining.
  • The antibody cross-reacts with mouse TBR1, making it suitable for studies using mouse cortical development models.
  • For ELISA, include appropriate negative and positive controls. The peptide immunogen can be used as a positive control for assay validation.
  • As with all antibody-based applications, empirical optimization of dilution and blocking conditions is recommended for each sample type.
