Biological Background: TBP Function and Localization
- TBP (TATA-box-binding protein) is a general transcription factor that plays a central role in initiating RNA polymerase II-dependent transcription. It functions as a subunit of the TFIID complex. Related references: PMID:33795473
- TBP specifically binds to TATA-box promoter elements, bending DNA and facilitating assembly of the pre-initiation complex (PIC), which includes RNA polymerase II and other general transcription factors. Related references: PMID:2194289, PMID:2363050, PMID:2374612
- It is a component of TFIID, a large complex comprising TBP and multiple TATA-binding protein-associated factors (TAFs TAF1–TAF13). Within TFIID, TBP forms the TFIID-A module together with TAF3 and TAF5. Related references: PMID:27007846, PMID:33795473
- TBP also participates in RNA polymerase III transcription via a BRF2-containing complex, and in RNA polymerase I transcription as part of the SL1/TIF-IB complex. Related references: PMID:26638071, PMID:15970593
- The protein localizes exclusively to the nucleus, where it engages with promoter DNA to regulate gene expression.
- TBP is widely expressed across tissues, with highest expression levels found in testis and ovary, reflecting its essential role in global transcription.
- Alternative splicing of the TBP gene yields multiple isoforms, and mutations in TBP are associated with neurodegenerative diseases such as spinocerebellar ataxia type 17 (SCA17).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (amino acids 1–100) of human TBP. Researchers should consider that different isoforms or post-translational modifications may affect antibody binding; validation in the specific experimental system is recommended.
- For Western blot, the predicted molecular weight of TBP is ~38 kDa. Multiple bands or slight shifts may be observed due to alternative splicing or modifications; include appropriate controls.
- The antibody is reported to cross-react with mouse and rat TBP, making it suitable for studies across these species. However, confirm cross-reactivity in your model by testing with known positive samples.
- For ELISA applications, optimize antibody concentration and assay conditions. Note that polyclonal antibodies may exhibit higher background; negative controls are essential.
- This antibody is supplied as a polyclonal antibody from rabbit, which typically recognizes multiple epitopes. This can be advantageous for denaturing assays like WB but may require careful blocking and washing steps.
