Biological Background: TAZ Function and Localization
- TAZ (WWTR1) is a WW domain-containing transcription regulator protein 1, also known as transcriptional coactivator with PDZ-binding motif, functioning as a downstream regulatory target in the Hippo signaling pathway. Related references: PMID:11118213, PMID:18227151, PMID:23911299
- It plays a pivotal role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis, acting as a proto-oncogene when dysregulated. Related references: PMID:11118213, PMID:18227151, PMID:23911299
- TAZ is phosphorylated and inactivated by LATS1/2 kinases within the Hippo kinase cascade, and it shuttles between the nucleus, cytoplasm, cell membrane, and tight junctions.
- It enhances PAX8 and NKX2-1/TTF1-dependent gene activation and, together with YAP1, regulates TGFB1-dependent SMAD2/3 nuclear accumulation, coupling SMADs to the transcriptional machinery. Related references: PMID:19010321, PMID:18568018
- TAZ is highly expressed in kidney, heart, placenta, and lung, and is also found in thyroid tissue.
- The protein contains WW domains, a coiled coil region, and an isopeptide bond, and undergoes ubiquitin conjugation; its molecular weight is approximately 44 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-270 of human TAZ (NP_056287.1), covering the N-terminal region; consider this epitope location when designing experiments that require specific domain recognition.
- This rabbit recombinant monoclonal antibody (PolymAb®) is suitable for western blotting (WB), immunohistochemistry on paraffin-embedded sections (IHC-P), immunoprecipitation (IP), and ELISA. Optimize dilution and incubation conditions for each application in your specific sample system.
- Cross-reactivity with human, mouse, and rat TAZ has been reported; validate reactivity in the relevant species and tissue type before quantitative assays.
