Biological Background: TAZ Function and Localization
- Encoded by the WWTR1 gene (also known as TAZ), WW domain-containing transcription regulator protein 1 is a transcriptional coactivator with a PDZ-binding motif.
- Transcriptional coactivator acting as a downstream regulatory target in the Hippo signaling pathway, playing a pivotal role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. Related references: PMID:11118213 PMID:18227151 PMID:23911299
- The Hippo pathway core kinase cascade involves STK3/MST2 and STK4/MST1 with SAV1 phosphorylating LATS1/2 with MOB1, subsequently phosphorylating and inactivating YAP1 and WWTR1/TAZ. Related references: PMID:18227151
- Enhances PAX8 and NKX2-1/TTF1-dependent gene activation. Related references: PMID:19010321
- In conjunction with YAP1, regulates TGFB1-dependent SMAD2 and SMAD3 nuclear accumulation and couples SMADs to the transcriptional machinery. Related references: PMID:18568018
- Regulates embryonic stem-cell self-renewal, promotes cell proliferation and epithelial-mesenchymal transition. Related references: PMID:18227151 PMID:18568018
- Subcellular localization includes nucleus, cytoplasm, cell membrane, and tight junctions.
- Highly expressed in kidney, heart, placenta, and lung; also expressed in thyroid tissue.
- Classified as a proto-oncogene; subject to phosphorylation and ubiquitination, with a coiled coil domain.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 300–400 of human TAZ. Epitope recognition may be conformation-independent; consider its suitability for denaturing and native applications.
- For Western blot (WB), the predicted molecular weight is approximately 44 kDa. Use nuclear/cytoplasmic extracts as appropriate and validate with positive and negative controls.
- Immunofluorescence/immunocytochemistry (IF/ICC) may require optimized fixation and permeabilization to detect nuclear and cytoplasmic staining. Co-staining with tight junction markers can confirm localization.
- For ELISA, consider pairing with a capture antibody if developing a sandwich assay, or use direct coating of the immunogen peptide for performance screening.
