Biological Background: TAX1BP3 Function and Localization

  • TAX1BP3, also known as Tax1-binding protein 3, TIP-1, or glutaminase-interacting protein 3, is a PDZ domain-containing adaptor protein that modulates multiple protein-protein interactions and signaling pathways.
  • Acts as a negative regulator of the canonical Wnt pathway by binding directly to beta-catenin (CTNNB1) and inhibiting its transcriptional co-activator function.
  • Competes with the scaffold protein LIN7A for binding to the inward-rectifier potassium channel KCNJ4, thereby promoting KCNJ4 internalization from the cell membrane.
  • Participates in Rho GTPase signaling and is reported to mediate activation of CDC42 by the high-risk HPV16 E6 oncoprotein, linking it to viral oncogenesis.
  • Localizes to the cytoplasm, nucleus, and cell membrane, consistent with its role in nucleocytoplasmic shuttling and membrane-associated signaling complexes.
  • Ubiquitously expressed in human tissues, with detection in brain, heart, kidney, lung, small intestine, and skeletal muscle, as well as in HeLa and other cell lines.
  • Post-translationally modified by acetylation and phosphorylation, as evidenced by large-scale proteomics studies. Also noted as a phosphoprotein and a reference proteome member.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1–100) of human TAX1BP3; consider verifying reactivity against full-length protein in the relevant species.
  • For Western blot, the expected molecular weight is ~14 kDa; a 15% SDS-PAGE gel or gradient gel is recommended for better resolution of low-molecular-weight proteins.
  • In ELISA, the antibody may be used for antigen capture or detection; optimization of coating conditions and antibody concentrations is advised using recombinant TAX1BP3 or cell lysates.
  • Cross-reactivity with mouse and rat is reported; sequence alignment shows high homology in the immunogen region, but validation in each species is suggested.
