Biological Background: Tau Function and Localization
- Tau (microtubule-associated protein tau) promotes microtubule assembly and stability and is involved in the establishment and maintenance of neuronal polarity. Related references: PMID:21985311
- The protein acts as a linker between axonal microtubules (via its C-terminus) and neural plasma membrane components (via its N-terminus). Related references: PMID:21985311, PMID:32961270
- Short tau isoforms allow cytoskeletal plasticity, whereas longer isoforms preferentially stabilize the cytoskeleton.
- Tau localizes to the cytoplasm, cytosol, cytoskeleton, and cell membrane; it is present in axons, dendrites, and can be secreted.
- Expressed in neurons: the PNS-tau isoform is found in the peripheral nervous system, while other isoforms are expressed in the central nervous system.
- Tau undergoes extensive post-translational modifications (phosphorylation, acetylation, methylation, glycation, ubiquitination) and is a key player in Alzheimer disease and other tauopathies.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 500-758) of human Tau, which includes the microtubule-binding repeat domains; consider this when interpreting isoform recognition.
- For Western blotting, the expected full-length band is ~79 kDa, but multiple isoforms between 45–68 kDa may also be detected depending on the sample.
- Brain tissue lysates are recommended for Western blot verification, as Tau is highly enriched in neurons.
- For immunohistochemical applications, validate antibody performance on relevant tissue sections and optimize antigen retrieval conditions as necessary.
- ELISA can be used to quantify Tau in biological fluids; however, appropriate controls and standard curves should be established.
