Biological Background: TAS2R10 Function and Localization
- TAS2R10 (Taste receptor type 2 member 10), also known as taste receptor family B member 2 (TRB2) or T2R10, is a G protein-coupled receptor involved in bitter taste perception.
- It functions as a gustducin-coupled strychnine receptor, signaling through PLCB2 and the calcium-regulated cation channel TRPM5 upon ligand binding.
- TAS2R10 is expressed in subsets of taste receptor cells located in the tongue and palate epithelium, and exclusively in gustducin-positive cells.
- The receptor is localized to the plasma membrane and possesses seven transmembrane domains, characteristic of GPCRs.
- Post-translational glycosylation is predicted, consistent with its annotation as a glycoprotein.
- TAS2R10 is implicated in sensory transduction of bitter compounds not only in the oral cavity but also in the gastrointestinal tract.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a sequence within amino acids 100–200 of human TAS2R10, predicted to be in an extracellular or intracellular loop region, making it suitable for detecting the target in WB and ELISA.
- For Western blotting, the predicted molecular weight is approximately 35 kDa; however, glycosylation may result in a higher apparent molecular weight.
- For ELISA, consider using this antibody for antigen capture or detection in indirect ELISA formats; optimize coating concentrations and detection conditions.
- Cross-reactivity with mouse and rat indicates epitope conservation; validate performance in the relevant species sample system as needed.
