Biological Background: TAP2 Function and Localization
- TAP2 (Antigen peptide transporter 2) is also known as ATP-binding cassette sub-family B member 3 (ABCB3), peptide supply factor 2 (PSF2), and really interesting new gene 11 protein (RING11). It belongs to the ABC transporter family and is a key component of the MHC class I antigen presentation pathway.
- As part of the TAP1-TAP2 heterodimer, TAP2 mediates unidirectional translocation of cytosolic peptide antigens into the endoplasmic reticulum (ER) for loading onto MHC class I molecules (PubMed:25377891, PubMed:25656091). Related references: PMID:25377891, PMID:25656091
- This translocation is ATP-dependent and involves a conformational cycle where the peptide binding site alternates between cytosol-facing (inward) and ER-lumen-facing (outward) states. ATP hydrolysis resets the transporter to the inward-facing state (PubMed:11274390, PubMed:25377891, PubMed:25656091). Related references: PMID:11274390, PMID:25377891, PMID:25656091
- TAP2 preferentially transports intracellular peptides of 8–13 amino acids with free N- and C-termini, and selects for peptides with a hydrophobic residue at position 3 and hydrophobic or charged residues at the C-terminal anchor (PubMed:11274390, PubMed:7500034, PubMed:9256420). Related references: PMID:11274390, PMID:7500034, PMID:9256420
- TAP2 functions as a scaffold within the peptide loading complex (PLC), critical for the assembly of peptide-MHC class I complexes and efficient antigen presentation (PubMed:1538751, PubMed:25377891, PubMed:26611325). Related references: PMID:1538751, PMID:25377891, PMID:26611325
- The protein localizes to the endoplasmic reticulum membrane, consistent with its role in importing peptides into the ER.
- Keywords associated with TAP2 include adaptive immunity, host-virus interaction, peptide transport, and protein transport, highlighting its role in immune surveillance and viral defense.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 450–686 of human TAP2, a region near the C-terminus. This sequence may show variability across species; consider confirming reactivity in your model system if using non-listed species.
- For Western blot (WB), a band corresponding to approximately 76 kDa is anticipated. Use appropriate loading controls and, if necessary, protease inhibitors to prevent degradation.
- This polyclonal antibody is specified for IHC-P, IF/ICC, and ELISA; optimal dilutions and protocols should be determined empirically. Consider antigen retrieval for IHC-P and fixation/permeabilization conditions for IF/ICC.
- For ELISA applications, pair with a suitable detection antibody and validate using recombinant TAP2 protein or cell lysates expressing the target.
