Biological Background: TAP1 Function and Localization

  • TAP1 (Antigen peptide transporter 1) is an ATP-binding cassette (ABC) transporter of the ABCB subfamily, also known as ABCB2, PSF1, RING4, and TAP1N.
  • It forms a heterodimer with TAP2 to mediate unidirectional translocation of cytosolic peptide antigens into the endoplasmic reticulum (ER) for loading onto MHC class I molecules, using ATP hydrolysis as an energy source. Related references: PMID:25377891 PMID:25656091 PMID:11274390
  • TAP1 preferentially transports peptides of 8-13 amino acids with free N- and C-termini, selecting for hydrophobic or charged residues at the C-terminal anchor and a hydrophobic residue at position 3; proline at position 2 is destabilizing. Related references: PMID:11274390 PMID:7500034 PMID:9256420
  • As part of the peptide loading complex (PLC), TAP1 acts as a molecular scaffold facilitating peptide-MHC class I assembly and antigen presentation. Related references: PMID:1538751 PMID:25377891 PMID:26611325
  • TAP1 localizes to the endoplasmic reticulum membrane, consistent with its role in peptide translocation.
  • It is highly expressed in professional antigen-presenting cells (monocytes, dendritic cells) and lymphocyte subsets (T cells, B cells, NK cells).
  • The protein features ATP-binding, magnesium-binding, and nucleotide-binding domains critical for its transport function, and is involved in adaptive immunity, host-virus interactions, and alternative initiation.

Experimental Guidance and Technical Tips

  • The immunogen was designed from the C-terminal region (aa 524-808), which lies in the cytoplasmic domain and is well-conserved between human and mouse, contributing to the observed cross-reactivity.
  • This antibody is suitable for WB and ELISA; consider optimizing dilution and detection conditions for your specific sample system.
  • For WB, the predicted molecular weight is approximately 81 kDa; verify band specificity with appropriate positive and negative controls.
