Biological Background: TAP1 Function and Localization
- TAP1 (Antigen peptide transporter 1) is an ATP-binding cassette (ABC) transporter of the ABCB subfamily, also known as ABCB2, PSF1, RING4, and TAP1N.
- It forms a heterodimer with TAP2 to mediate unidirectional translocation of cytosolic peptide antigens into the endoplasmic reticulum (ER) for loading onto MHC class I molecules, using ATP hydrolysis as an energy source. Related references: PMID:25377891 PMID:25656091 PMID:11274390
- TAP1 preferentially transports peptides of 8-13 amino acids with free N- and C-termini, selecting for hydrophobic or charged residues at the C-terminal anchor and a hydrophobic residue at position 3; proline at position 2 is destabilizing.
