Biological Background: TAOK2 Function and Localization
- Encodes serine/threonine-protein kinase TAO2, also known as prostate-derived STE20-like kinase 1 (PSK1) and thousand and one amino acid protein kinase 2. Isoforms 1 and 2 are produced by alternative splicing.
- Functions in stress-activated MAPK cascades: phosphorylates MAP2K3 and MAP2K6 to activate p38/MAPK14, and participates in DNA damage checkpoint control at the G2/M transition.
- Isoform 1 induces apoptotic morphological changes (cell contraction, membrane blebbing, apoptotic body formation) via MAPK8/JNK and mitochondrial CASP9 pathways; localizes to the nucleus when C-terminally truncated.
- Isoform 1 binds microtubules and modulates their organization and stability independently of kinase activity. Prevents MAP3K7-mediated NF-kappa-B activation but not MAPK8/JNK activation.
- Isoform 2 is required for PCDH8 endocytosis. Upon homophilic PCDH8 interactions, isoform 2 phosphorylates p38/MAPK14, leading to reciprocal phosphorylation and internalization of PCDH8 and CDH2.
- Subcellular locations include cytoplasmic vesicle membrane, cytoplasm, cytoskeleton, nucleus, and cell projection dendrites. Ubiquitously expressed, with higher levels in testis and brain.
- Belongs to the protein kinase superfamily and contains signature motifs for ATP binding, magnesium coordination, and phosphotransferase activity.
