Biological Background: Talin-1 Function and Localization
- Talin-1 is a high molecular weight cytoskeletal protein encoded by the TLN1 gene (also known as KIAA1027, TLN) in humans.
- It is concentrated at regions of cell-substratum and cell-cell contacts, serving as a critical linker between the actin cytoskeleton and integrin receptors at focal adhesions.
- Talin-1 plays a key role in mechanically coupling the cytoskeleton to the extracellular matrix, facilitating signal transduction and cell adhesion.
- In collaboration with KANK1, Talin-1 co-organizes cortical microtubule stabilizing complexes (CMSCs) at focal adhesion rims to regulate microtubule-actin crosstalk.
- Subcellularly, Talin-1 localizes to the cytoplasm, cytoskeleton, cell surface, cell junctions, focal adhesions, and cell projections such as ruffle membranes.
- Talin-1 is expressed in many tissues, with particularly high expression in skin and pancreas, and robust expression in kidney cortex, endocervix, testis, pituitary, liver, and spleen.
- The protein undergoes multiple post-translational modifications including acetylation and phosphorylation, and is involved in host-virus interactions.
